What is Daurismo?

Daurismo, also known as glasdegib (glassdigib), belongs to the class of drugs called hedgehog pathway inhibitors. Glasdegib blocks the action of an enzyme that tells cancer cells to multiply. It helps to stop or slow down the spreading of cancerous cells. Daurismo and cytarabine are used as the first treatments for acute leukemia in patients over 75, in addition to other medications that cannot be prescribed. Daurismo hasn't been tested on people who have severe liver or kidney disease.

Warnings

Daurismo should not be used by women who are or may become pregnant. If either the mother or the father are taking glasdegib, there is a very high chance that the baby will be born with severe birth defects or die. Use birth-control. You must take a pregnancy test and have it come back negative within seven days of starting treatment. You must use an effective form of birth control while you are on Daurismo, and at least for 30 days following your last dose. Speak to your doctor regarding birth control methods that work best for you. You should be aware that glasdegib can cause harm to an unborn child if you have a partner, male or female, who has the potential for pregnancy. Even if you've had a vasectomie (surgery performed to stop sperm from leaving the body, causing pregnancy), use a condom for 30 days following your final Daurismo dose. Don't donate any semen while you are receiving treatment or for 30 days following your final dose. Call your doctor immediately if you think your contraception has failed or if you think your partner or you may be pregnant. Don't donate blood during the 30 days following your Daurismo treatment.

Before you take this drug

Tell your doctor about any of the following to ensure that Daurismo will be safe for you:

  • Heart problems
  • Long QT Syndrome
  • An electrolyte balance (such as a low potassium or magnesium level in the blood);
  • Muscle pain or muscle spasms
  • Kidney disease
  • Liver disease

Within 7 days after starting Daurismo treatment, you will be required to perform a pregnancy check. Can harm an unborn child. A pregnancy test is required to confirm that you are not expecting. Daurismo should be used with effective birth control for 30 days following your last dosage.As soon as you discover you've become pregnant, inform your physician immediately. You should not breastfeed for 30 days following your last dose of this medication. Daurismo users should continue to use condoms for 30 days following treatment. This includes after a vasectomy. You may find it harder to get pregnant if you use this medication.

How to take Daurismo?

Read all the instructions and directions provided by your doctor on the prescription label.Sometimes your doctor will adjust your dosage. Daurismo can be given as a 28-day treatment cycle. Daurismo must be taken orally. You will be prescribed this medication by your doctor. Medicine should be taken at approximately the same time daily. Do not chew or crush the tablet. Do not continue taking Daurismo if you vomit soon after taking it. Do not miss your scheduled dose. Electrocardiograms (also called EKGs) are used to check the heart's function. Your cancer treatment may be delayed, depending on the test results. Keep away from heat and moisture at room temperature.

Details on dosage

Adult dose of Daurismo:

In the absence of unacceptable toxicities or disease control, 100 mg once daily on days one to 28 with 20 mg cytarabine subcutaneously twice a day from days one to ten of every 28-day cycle; for those patients who do not have unacceptable toxicities, treatment should be continued for at least 6 cycles in order to give time to clinical response.
Use: In combination with low-dose cytarabine for newly diagnosed acute myeloid leukemia in adults who are at least 75 years of age or have comorbidities that preclude the use of intensive chemotherapy.

The Usual Geriatric Daurismo Dose for Acute Myeloid Leukemia:

Age 75 and older
In the absence of unacceptable toxicities or loss of disease, 100 mg once daily on days 1–28 with 20 mg of cytarabine subcutaneously twice a day from days 1–10 of each cycle of 28 days is recommended for those patients who do not have unacceptable toxicities or no clinical response to treatment.
Use this drug in conjunction with low-dose Cytarabine for newly diagnosed acute myeloid leukemia in adults who are at least 75 years of age or have comorbidities that preclude the use of intensive chemotherapy.

What happens if I miss the dose?

If you miss a dose, take it immediately.Skip the dose missed if the next dose is in 12 hours or less. You should not take two doses in a 12-hour period.

What happens if I overdose?

Call 1-800-222-1222 immediately if you suspect being poisoned and/or seek immediate medical advice. The symptoms of an overdose may include fatigue, nausea, and vomiting. You might also feel dizzy, like you're about to pass out, or thirsty.

What should be avoided?

Don't donate blood while you are taking Daurismo and at least 30 days after your last dose. Don't donate sperm while taking Daurismo and at least 30 days after the last dose.

Side effects of Daurismo

If you experience any of the following symptoms of an allergy to Daurismo, seek immediate medical attention: Hives, difficulty breathing, swelling in your mouth, face, lips, or throat

Daurismo may cause serious side effects. If you experience:

  • Shortness of breath and dizziness, or feeling like you may pass out, are all signs that your heart is beating too fast.
  • Easy bruising or bleeding; purple or red patches under the skin
  • Muscle pain, spasms, or any other new or worsening symptoms
  • Low blood counts: fever; chills; easy bruising and bleeding. Pale skin. Cold hands and feet. Feeling lightheaded.

If you experience certain side effects, your cancer treatment may be permanently stopped or delayed.

Common Daurismo side effects may include:

  • Bleeding;
  • Fever and low blood cell counts
  • Changes in your taste buds;
  • Pain or sores in the mouth or throat
  • Loss of appetite, constipation, and nausea
  • Rash;
  • Muscle pain;
  • You may experience swelling in your legs or arms.
  • Feeling tired
  • Trouble breathing

Interaction with other drug

It is sometimes not safe to take certain medications at the same time. Some medicines can cause your blood level of another drug to increase, increasing side effects and making the medicine less effective. Daurismo may cause serious heart problems if taken with certain medications for asthma, diabetes, high blood pressure, depression, mental illnesses, HIV, malaria, or cancer. Other medications, such as vitamins and herbal remedies, may also interact with glasdegib. Inform your doctor of all the other medications you take.
