What is Loncastuximab Tesirine?

Loncastuximab-tesirine can be used to treat large lymphomas when at least two other cancer therapies have failed or stopped working.This medication guide does not list all possible uses of Loncastuximab Tesirine.

Side effects of Loncastuximab tesirine

Seek immediate medical care if you have any of these symptoms: difficulty breathing, swelling in your face, lips, throat, or tongue.

Loncastuximab tesirine my cause serious side effects. If you experience:

  • Skin rash or new skin;
  • Chest pain or difficulty breathing.
  • Fluid buildup around or in the lungs causes pain when you breathe. Feeling short of breath, lie down. Wheezing. Cough. Foamy mucus. Cold and clammy. Anxiety.
  • Low blood cell counts: fever; chills; tiredness; easy bruising and bleeding; pale or cold skin, hands or feet; or feeling lightheaded or short of breath.
  • Signs of a serious virus include flu symptoms, aches and pains, headaches, or difficulty breathing.

Some of the most common side effects associated with loncastuximab Tesirine include:

  • Tiredness or weakness
  • Rash;
  • Swelling;
  • Nausea;
  • Joint or muscle pain
  • Increase in blood sugar
  • Changes in blood or laboratory tests

Warnings

You could get serious or even fatal infections. You should call your doctor if there are any of the following symptoms: fever, chills or flu symptoms (coughing, fatigue, weakness, or body aches), headaches, breathing problems, or wounds that have pain, swelling, redness, or warmth.

Before you take this drug

Tell your doctor about any of the following:

  • Liver problems
  • An infection is a serious illness.

Loncastuximab Tesirine can cause harm to an unborn child if either the mother or father uses it.

  • You may need to take a pregnancy test if you're a woman. Use birth control during treatment and for 9 months following your last dose.
  • Use birth control if you're a man and your partner can get pregnant. Continue to use birth control at least six months after the last dose.
  • If you fall pregnant, inform your doctor as soon as possible.

This medicine may make it harder for men to get women pregnant. Use birth control to avoid pregnancy, as the medicine may harm an unborn child.

Do not breastfeed while taking this medicine or for 3 months following your last dose.

How to take loncastuximab Tesirine?

Loncastuximab Tesirine is usually injected by a healthcare professional into a vein once every three weeks.Other medications may be prescribed to prevent severe side effects. Continue to use these medications as prescribed by your doctor.You could get more serious infections, or even fatal ones. You may need to undergo frequent medical tests, and your doses could be delayed depending on the results.

Details on dosage

Adult dose for lymphoma:

For the first two cycles, 0.15 mg/kg intravenously over 30 minutes on Day 1 (every three weeks), then 0.075 mg/kg intravenously over 30 minutes every 3 weeks.
Comments:
Premedicate with Dexamethasone 2 mg orally or IV twice a day, starting the day before taking this drug.
If dexamethasone is not administered the day prior to this drug, it should be started at least two hours before administration.
Use: To treat adult patients with relapsed large B-cell lymphoma or refractory lymphoma following 2 or more systemic therapies, including diffusive large B-cell (DLBCL), not otherwise specified. DLBCL that arises from low-grade lymphoma

What happens if I miss the dose?

If you missed your appointment to receive your loncastuximab-tesirine injection, call your doctor.

What happens if I overdose?

In a medical environment, an overdose is treated immediately.

What should be avoided?

You may get sunburned easier if you take Loncastuximab Tesirine. Avoid tanning beds or sunlight. When you're outdoors, wear protective clothing and use sunblock (SPF 30 or higher).

Interaction with other drug

Other drugs can affect loncastuximab and tesirine. These include prescription and over-the-counter medicines, vitamins, and herbal products. Inform your doctor of all the medicines you are taking and those that you stop or start using.
