What is Entrectinib?

Entrectinib is a treatment for adult patients to manage the non-small cell lung tumor that has spread to various regions within the human organs (metastatic).Entrectinib may also be prescribed to adult patients and children who are less than 12 years old with solid tumors that have grown or are not completely removed by surgery, or if other treatment options have not worked or aren't viable alternatives.Entrectinib is only used when cancer is caused by a particular abnormal gene. Your physician will test you to determine the presence of this particular gene.Entrectinib is also used for reasons not mentioned in this medication guide.

Side effects of Entrectinib

See a doctor immediately. Get medical attention immediately if you notice symptoms or warning signs of an allergic response, such as difficulty breathing, hives, and swelling of your lips, face, and throat.

Entrectinib could cause serious adverse reactions. Consult your physician immediately in the event you suffer from:

  • Bone pain or issues with movement;
  • Rapid or pounding heartbeats, the chest fluttering with excitement, shortness of breath, and a sudden feeling of dizziness (like you're about to pass out);
  • Nausea, vomiting, and loss of appetite; upper stomach pain on the right side;
  • Swelling in your lower legs, rapid weight gain;
  • Cough, wheezing, breathlessness, and difficulty breathing while lying down
  • Confusion, hallucinations, or problems in concentration or memory;
  • Dizziness, mood changes, trouble sleeping,
  • Double vision or blurred vision makes you more sensitive to light or seeing flashes of light, or "floaters" in your vision.

The treatment you receive for cancer could be delayed or discontinued permanently in the event of certain negative side effects.

Common negative side effects of Entrectinib can include:

  • Shifts in mental state;
  • Vision change;
  • Nausea, vomiting, diarrhea, constipation;
  • Fever, cough;
  • Dizziness, tiredness;
  • Swelling in your lower legs, rapid weight gain;
  • Muscles or joint discomfort;
  • Trouble breathing
  • Shifts in your perception of touch or taste.

Cautions

Entrectinib can result in new or worsening manifestations of constricted heart disease. Contact your physician immediately when you notice swelling of your lower legs, weight loss that is fast coughing, wheezing, breathing problems, or difficulty breathing when lying on your back.

Prior to using this drug

Speak to your doctor if you were ever diagnosed with:

  • Gout;
  • Heart-related problems;
  • Long qt syndrome (in the case of you as well as a relative);
  • Kidney or liver disease;
    a disorder of the nervous system;
  • Issues with your eyes or problems with your eyesight.

It is possible that you will need to take a pregnancy test that is negative prior to commencing this treatment.Entrectinib could harm a newborn baby as well as cause birth defects when either the father or mother is taking the drug.

  • If you're female, do not use entrectinib while you are pregnant. Utilize efficient contraception to avoid pregnancy while taking this medicine and for a minimum of 5 weeks following your last dose.

How do I take entrectinib?

Follow the directions on the label of your prescription and read the medication guide or instructions sheets. The doctor might alter your dosage. Make sure you take the medicine precisely as directed.Take your daily dosage either alone or with meals.Take the capsule in its entirety and don't crush or chew on it, break it, or break it open.If you have a stomach ache within a few minutes of taking entrectinib, take a second dose.Place it in a cool, dry place free of heat and moisture.Don't alter the dose or stop taking entrectinib unless your doctor's guidance is given.

Info on dosage

Usual Adult Dose for Non-Small Cell Lung Cancer:

600 mg taken orally every day up to the point of being unacceptably toxic
Use: For adults who have metastatic noncancers of the non-small cells lung cancer (NSCLC) who have tumors that are ROS1-positive

Usual Adult Dose for Solid Tumors:

600 mg taken orally every day up to the point of being unacceptably toxic
Uses: For patients suffering from solid tumors that are characterized by neurotrophic tyrosine receptor kinase (NTRK) gene-fusion and do not have an acquired resistance mutation that is known to be metastatic or in which surgical resection could cause significant morbidity and have deteriorated after treatment or do not have an alternative treatment.

Usual Pediatric Dose for Solid Tumors:

12-year-olds and older:
Body surface area (BSA) higher than 1.5 millimeters 600 mg taken orally, every day
BSA 1.11 up to 1.5 m2 500 mg, orally, twice every day
--BSA 0.91 to 1.1 M2 400 mg, orally every day
Use: for children 12 years of age or older who have solid tumors with neurotrophic tyrosine kinase (NTRK) gene-fusion but without any known mutation that causes resistance and is metastatic, or in cases where surgical resection could cause severe morbidity and have worsened since treatment or do not have an alternative treatment.

What happens if I miss the dose?

Utilize the medicine as quickly as you are able, but take a break from any missed doses if the coming dose falls to be taken in under 12 hours. Do not take two doses at once.

Avoid this

Avoid driving or engaging in hazardous activities until you understand how the effects of entrectinib affect your body. Your reaction could be affected.Grapefruit could interfere with entrectinib and cause adverse negative side effects. Beware of using grapefruit-related products.

Interaction with other drug

It is sometimes not recommended to take certain medicines in combination. Certain drugs may affect the blood levels and the other medications that you take, which can cause more side effects or make the medication less effective.Other medications can influence entrectinib. This includes medications that are prescribed and available over the counter, vitamins, and herbal products. Discuss with your doctor all the medicines you are currently taking and any medication you begin or stop taking.
