What is Amifampridine?

Amifampridine is a treatment for the myasthenic condition Lambert-Eaton in adults.Amifampridine is also used for reasons not mentioned in this guideline.

Side effects of Amifampridine

Contact a medical professional immediately. Get medical attention immediately if you notice symptoms or warning signs of an allergic response, such as difficulty breathing, hives, or swelling of your lips, face, or tongue.Amifampridine can trigger seizures even if you have never experienced a seizure before.\Stop using amifampridine immediately, and call your doctor immediately when you're experiencing seizures.

Common side effects of amifampridine can include:

  • Tingling sensation in your feet, hands, face, or any other area of your body.
  • Nausea, nausea, stomach pain, diarrhoea,
  • Headache, back pain;
  • Elevated blood pressure
  • Abnormal tests of liver function;
  • Muscle spasms
  • Symptoms of the cold, like nasal stuffiness, sneezing, and sore throats.

Warnings

Amifampridine can trigger seizures even if you've not had any before. Stop taking amifampridine and contact your doctor if you are experiencing seizures.

Prior to use this drug

Amifampridine should not be used in the event that you are allergic to it, or:

  • You've had seizures in the past.
  • You are sensitive to the chemical dalfampridine.

Consult your doctor If you've ever had:

  • A seizure;
  • The liver condition;
  • Kidney disease
  • If you take another form of amifampridine that you can purchase from the pharmacy that compounded it (3,4-diaminopyridine, also known as 3,4-DAP),

Inform your doctor if you are nursing or pregnant.

How to take Amifampridine?

Follow the directions on the prescription label and go through all medication guides or instructions. The doctor might alter your dosage. Follow the medication exactly as prescribed.Amifampridine is a drug that can be taken either with or without food.Don't take more than two tablets at once. Don't take more than 8 tablets within 24 hours.The amifampridine tablet is able to be split in two if required.

Details on dosage

Usual Adult Dose for Lambert-Eaton Myasthenic Syndrome:

Initial dosage: 15 to 30 milligrammes daily, in split doses (3–4 times a day).
Maximum single dose: 20 mg
Maximum daily dose: 80 mg/day
Comments:
The dose can be increased by 5 mg over the course of 3–4 days.
If a dose is missed, the patient should not take more than one dose or double it.
Use: To treat Lambert-Eaton myasthenic disorder (LEMS)

Usual Paediatric Dose for Lambert-Eaton Myasthenic Syndrome:

From 6 to 17 years old, weighing less than 45kg:
Initial dosage: 7.5 mg to 15 mg once daily in doses that are divided (2 or 3 times a day).
Maximum single dose: 15 mg
Maximum daily dose: 50 mg
Comments:
The dose can be increased in 2.5 mg increments from 5 mg to 2.5 mg and divided into 5 doses per day.
Dosage should be increased depending on the response of patients and their tolerance.
A suspension is prepared for patients who require a dose smaller than 5 mg per increment.
Between 6 and 17 year old, weighing up to at least 45kg:Initial dosage Between 15 and 30 mg in split portions (2 or 3 times per day)
Maximum single dose: 30 m
Maximum daily dose: 100 mg
Comments:
Dose can be increased by 5 mg–10 mg increments and divided into 5 doses per day.
Dosage should be increased based on clinical response and tolerance.
Use: To treat Lambert-Eaton myasthenic disorder (LEMS)
